![A. Isolation of Chloroplasts
1. Devein 10g of fresh papaya leaves and wash with distilled water.
2. Place the leaves and 50 mL cold phosphate buffered saline (PBS, 0.2 M
phosphate buffer, pH 7.0 and 0.35 M NaCl solution), in an ice-cold blender.
3. Blend at high speed for 30 sec, then cool the jar in an ice bucket for 30 sec. Do this
three times.
4. Strain the extract through two layers of cheesecloth into an ice-cold beaker.
5. Transfer the suspension into two cold 50-mL centrifuge tubes ensuring that they
are of equal weight.
6. Centrifuge at 200 xg for 5 min.
7. Decant the supernate into two clean 50-mL centrifuge tubes and discard the pellet.
8. Centrifuge at 1400 xg for 15 min. Discard the supernatant and resuspend the pellet
by adding 5_mL PBS to each tube. Collect all suspensions in a small beaker
immersed in an ice bath. (Note: Compute for rpm value of the angular speeds
