Cytosolic protein lack N terminal signal peptide. Translation completes on free cytosolic ribosomes. They show post translational targeting. 

Protein targeting in nucleus : 

  • Protein is transported in folded form. 
  • Require Nuclear Localization Signal/ NLS 
  • NLS must be exposed outside  

Importin can recognize cytosolic protein with NLS and bind with it. Importin  along with cytosolic protein then transported into the nucleus through nuclear pore complex in ATP dependent manner. Inside the nucleus , protein dissociated from importin.Ran-GTP nxt bind with importin  and goes outside of the nucleus. In cytosol , RanGTP convert to Ran GDP and dissociated from importin . RanGDP goes to inside the nucleus and importin stays outside .
